RESUMO

The objective of this study was to report the occurrence of Mycobacterium avium subspecies paratuberculosis (MAP) in dairy goats, via description of their clinical presentation, histopathological findings, and molecular identification of the infectious agent. Screening was performed using IS900 real-time PCR (qPCR) in milk samples from 179 properties in the semiarid of Northeast region of Brazil. Pooled milk samples from all lactating goats from processing plants were submitted to molecular diagnosis. One property had a positive result at qPCR. The production unit which had the positive sample for MAP was located, and an on-site visit to this property was performed to collect individual milk samples, seven of which tested MAP positive by IS900 qPCR. With permission from the owner, two goats (Animal 1 was positive and Animal 2 was negative on first qPCR for MAP) were acquired and euthanized. Animals 1 and 2 had milk and portions of the duodenum, ileum, colon, and mesenteric lymph nodes positive at qPCR for MAP. Animal 1 also had MAP DNA detected in part of the jejunum and cecum. In animal 2, the ileocecal valve tested positive. MAP was not detected in the blood or feces of either animal; however; it was confirmed for the association of clinical findings, histopathology, and qPCR. The gene IS900 from the positive samples were sequenced and showed a 99% similarity with MAP. The MAP was identified for the first time in the goat milk and tissues in the semiarid region of Northeast Brazil.

RESUMO

OBJECTIVE: The objective of this study was to evaluate the hemostatic activity of the sap from Jatropha mollissima (Pohl) Baill. in rats. MATERIALS AND METHODS: Twenty-four Wistar rats were randomized into four groups (n = 6): the JM25 and JM40 groups were treated with ethanolic extract from the sap of J. mollissima, in a concentration of 25 and 40 mg·mL1, respectively; the MO group was treated with Monsel's solution and the control group SC with a 0.9% sodium chloride solution. STATISTICAL ANALYSIS: Data were submitted to the Kurskal-Wallis' test, followed by Dunn's post hoc (p < 0.05). RESULTS: There was a significant reduction in the bleeding time of the group from the JM25 extract (p = 0.001) when compared with MO and SC. There were no statistically significant differences between groups JM25 and JM40 (p > 0.05). The JM25 group did not present rebleeding, a result significantly different from the MO group (p = 0.001). Monsel's solution showed significant bleeding, six times greater than the control group SC. CONCLUSION: The J. mollissima extract, in the concentration of 25 mg·mL1, showed the highest hemostatic efficiency and was found to be a promising biomaterial for the elaboration of a hemostatic product.

RESUMO

ABSTRACT: We described a case of systemic infection by Coccidioides sp. in a dog. An adult, mixed breed, free-ranging male dog presented with clinical signs that included apathy, cachexia, anorexia, limited mobility with sternal recumbency, bilateral mucopurulent ocular discharge, dyspnoea, pulmonary crepitation, erosive and nodular lesions on the skin, and swelling and stiffness of the left tibiotarsal joint. The dog was submitted to a postmortem examination. Grossly, there were multiple yellow to white nodules in various organs. Histologically, the lesions were characterized as pyogranulomatous inflammation associated with fungal spherules morphologically consistent with Coccidioides sp. The dog was concomitantly diagnosed with undifferentiated sarcoma affecting the skin, lymph nodes, liver, and testicles. The diagnosis of coccidioidomycosis was made based on the histologic changes associated with morphotintorial features and positive immunolabeling of organisms with anti-Coccidioides immunohistochemistry. This case demonstrated that Coccidioides sp. can infect dogs that inhabit urban centers in the semiarid region of Northeastern Brazil, likely due to exposure to dust from contaminated environments.

The main clinical, anatomopathological, and molecular aspects of the infection by Leishmania infantum are described in two cats with multicentric cutaneous, nodular, and ulcerated lesions. The animals were submitted to a clinical examination, followed by serological, molecular and parasitological exams, with culture and isolation of the parasite, and subsequent isoenzymatic characterization. The animals were euthanized and necropsied. Case 1 was an adult, female, mixed-bred stray cat. Case 2 was an adult, male, mixed-bred and domiciled cat. Both were positive for the presence of anti-L. infantum antibodies. In the cytology of the cutaneous nodules and lymph nodes, amastigote forms of Leishmania spp. could be visualized, free and in the interior of the macrophages. In the histopathology, the lesions were characterized by nodular granulomatous and/or ulcerative dermatitis, associated to amastigote forms of Leishmania spp. By means of the polymerase chain reaction, the sequence of the L. infantum kDNA minicircle was amplified. It is concluded that the infection by L. infantum occurs in cats in the State of Paraíba, Northeast region of Brazil and the need to understand the immunological profile of the visceral leishmaniasis in the feline population is highlighted with aimed at the control measures in public health.

RESUMO

A 7-year-old, white, domestic short hair, female cat was presented with an aural hematoma of the right pinna, which had been partially resected a year previously. A 3 × 4 cm mass, macroscopically similar to an auricular hematoma, was visible on the convex surface and a smaller vascular lesion was present on the ear margin. Cytological examination of the smaller mass was suggestive of hemangiosarcoma, and a diagnosis confirmed by histopathologic and immunohistochemical tests. Complete pinnectomy with a wide margin was performed and during the 3-year follow-up, the cat had no local recurrence or metastasis. We conclude that hemangiosarcoma should be considered as a potential differential diagnosis in white-coated cats with an unusual clinical presentation of aural hematoma.

RESUMO

Even nearly forty years after the eradication of smallpox, members of the Poxviridae family continue to be the focus of an increasing number of studies. Among these studies, prominently stands vaccinia virus, an orthopoxvirus that is associated with bovine vaccinia outbreaks. Although more frequently associated with infections in cattle and humans, the host range of vaccinia virus is not restricted only to these hosts. There are several instances of molecular and serological evidence of circulation of vaccinia virus among wildlife species. In addition, viral isolation has confirmed a broad spectrum of vaccinia virus hosts. In this report, we provide a brief update on the host range of Brazilian vaccinia virus, and present a case description of an outbreak in domestic buffalo calves from Northeastern Brazil that corroborates previous serological and molecular studies. Furthermore, in the present study, vaccinia virus has been isolated for the first time in buffaloes, and referred to as vaccinia virus Pernambuco (VACV-PE). Phylogenetic reconstruction was based on A56R clustered VACV-PE with vaccinia virus isolates belonging to group 1 Brazilian vaccinia virus. Furthermore, the vaccinia virus genome was detected in the milk of a lactating cow, which thereby revealed a pathway for future studies on the possible impact of vaccinia virus on buffalo milk and milk products. Taken together, these results provide the first description of clinical disease caused by vaccinia virus in buffaloes in South America. They also raise new questions about the chain of transmission of this virus.

RESUMO

In areas where human tuberculosis and bovine tuberculosis coexist, differentiation between M. bovis and M. tuberculosis is important for monitoring the spread of M. bovis among cattle and from cattle to humans. The objective of this study was to isolate and identify M. bovis in bovines with positive diagnosis identified on tuberculin test in the State of Paraíba, Northeastern Brazil. Thirty-two bovines that tested positive in the comparative tuberculin test were used, from which samples of any organ with lesions suggestive of tuberculosis were collected, as well as lymph nodes, when no gross lesions were observed. Samples were submitted to histopathological exam, mycobacterial culture, Ziehl-Neelsen staining and molecular diagnosis. Twenty-one (65.6%) animals presented lesions suggestive of tuberculosis. As to body region 77.7% of lesions were found in the thoracic cavity, 12.4% in the head and 9.9% in the abdominal cavity. Among 55 samples submitted to mycobacterial culture, mycobacteria were isolated in 31 (56.4%), being 13 (41.9%) identified as M. bovis and 18 (58.1%) as Mycobacterium spp. Conclusion is that isolation and identification of M. bovis and Mycobacterium spp. in cattle suggests that humans are exposed to the risk of infection. This reinforces the need for intensification and optimization of prevention and control measures foreseen in the Brazilian National Program for the Control and Eradication of Bovine Brucellosis and Tuberculosis. Mycobacteria isolation and identification surveys are, therefore, encouraged in other Northeastern states.(AU)

ABSTRACT: The aim of this study was to assess systemic and neurotoxic changes following an epidural administration of meloxicamin to rabbits. Twelve adult rabbits four males and eight females; average mass, 1.9 ± 0.1kg were randomly divided into two groups: a control group (GC), which received a single dose of 0.9% NaCl epidurally in a volume of 0.3mL kg-1and a meloxicam group (GM), which received 0.2mg kg-1 meloxicam epidurally along with 0.9% NaCl in a total volume of 0.3mL kg-1. Heart rate, respiratory rate, body temperature, and neurological abnormalities were assessed prior to administration of anesthesia (H0), 1, 2, 3, 6, 12, and 24h following epidural puncture (H1, H2, H3, H6, H12, and H24, respectively), and every 24h afterward for 10 days after epidural puncture (D2, D3, D4, D5, D6, D7, D8, D9, and D10). The surface temperature of lumbosacral region was also measured at H0, H1, H6, H12, H24, D5 and D10. Three animals from each group were euthanized on days 15 and 30 after epidural puncture to assess possible spinal injuries. Variances observed in physiological parameters were not suggestive of adverse effects of meloxicam, as all were within the reference standards, and there were no physical or behavioral changes observed. Neurological function was similar between groups, with only difference between baseline values and values 1h after epidural administration in both groups. There were no histopathological changes in the GM group, and only one animal showed discrete lymphocytic infiltrate. Epidural lumbosacral administration of meloxicam at a dose of 0.2mg kg-1 caused no significant systemic or neurotoxic effects in rabbits.

With the objective to determine the epidemiological aspects of conidiobolomycosis and rhinofacial pythiosis in sheep, a case-control study was conducted in 27 farms (16 cases and 11 controls) in the states of Paraíba and Rio Grande do Norte. In nine (56.3%) farms only one outbreak was observed and in seven, two outbreaks occurred, totaling 23 outbreaks. In 20 farms only cases of conidiobolomycosis occurred and in three farms cases of conidiobolomicosis occurred together with cases of pythiosis. In conidiobolomycosis, morbidity ranged from 0.7% to 73.3% and lethality rate was 100%. Thirteen outbreaks (56.3%) occurred in the rainy season (January to May), six (26.1%) in the dry season (June to December) and in four it was not informed. In all farms with cases, sheep had access to ponds for the whole year or for periods of 2-8 months. In the univariate analysis of risk, the variables associated with the occurrence of conidiobolomycosis were: animal grazing on the borders of the ponds; presence of vegetable matter consisting mainly of aquatic plants in the pond borders; period of grazing on the pond borders; high grazing pressure; and predominant breed of sheep, being the Santa Inês breed the most affected. It was concluded that the combination of factors such as continuous grazing in wetlands with decomposed vegetable matter and soil rich in organic matter exposes the animals to Conidiobolus spp. and predisposes to the occurrence of conidiobolomycosis. The establishment of pastures for cutting at the borders of the ponds can be an effective measure to control conidiobolomycosis.

RESUMO

The aim of this study was to determine the duration of the resistance after the end of the ingestion of non-toxic doses of Crotalaria retusa seeds. Ten sheep were divided into 3 groups of 3 animals each and a control group with 1 sheep. To induce resistance, sheep in groups 1, 2 and 3 received 20 daily doses of 2g kg-1 of C. retusa seeds, followed by 7 daily doses of 4g kg-1. To determine the duration of resistance the sheep in groups 1, 2 and 3 were challenged orally, 3, 7 and 15 days, respectively, after receiving the last dose of 4g kg-1, with a single dose of 5g kg-1. Sheep of groups 1 and 2 did not develop overt signs of poisoning. Two sheep of group 3, challenged 15 days after the end of the resistance induction period, showed signs of acute poisoning and died. These results suggest that the induced resistance of sheep to acute poisoning by C. retusa seeds is of short duration, from 7 to 15 days.

A case of lissencephaly and cerebellar hypoplasia was observed in a 30-day-old goat. The goat presented with sternal recumbence, absence of a menace response, intention tremors, ataxia, and nystagmus. The goat was euthanized and necropsied after having been hospitalised for eleven days. At necropsy, the surface of the brain was found to be smooth, the cerebral sulci and gyri were absent, and the cerebellum was reduced in size. Histologically, the grey matter and white matter were thicker and thinner than normal in cortices, respectively. The neurons were randomly arranged in the grey matter. In the cerebellum, the layers were disorganised, and cells were heterotopics. The histologic and gross lesions observed in this animal are characteristic of lissencephaly associated with cerebellar hypoplasia. The presence of a single goat affected suggests that the malformation was not of infectious origin and because lissencephaly is a malformation not previously described in goats, it is unlikely this case was inherited.

In this study, we investigated the presence of gastrointestinal helminths in 97 captive birds (Psittaciformes and Accipitriformes) necropsied between June and December 2011 in the state of Paraiba, Brazil. Forty-three birds were infected. Psittaciformes were infected by nematode Ascaridia hermaphrodita (97.6%) and cestode Raillietina sp. (2.4%). A. hermaphrodita was found in all species of parrots and Raillietina sp. was found only in Amazona aestiva. A. hermaphrodita was the cause of death, by intestinal obstruction, in 14 of the 40 birds investigated. Accipitriformes were infected by nematode Synhimantus (Synhimantus) rectus (100%) and acantocephalan Centrorhynchus tumidulus (50%). In Brazil, Diopsittaca nobilis and A. aestiva are reported for the first time as hosts of A. hermaphrodita and Raillietina sp., respectively. We concluded that Psittaciformes and Accipitriformes in captivity are affected by nematodes, cestodes and acanthocephalans and that implementation of control measures is essential.

RESUMO

We report exogenous pigmentation in sheep grazing in native pastures in northeastern Brazil. The sheep carcasses from a farm were condemned at the slaughterhouse due to pigmentation of the carcasses and viscera. In visits to the farm, bluish-purple pigmentation of the mucosa was observed in the sheep. In two necropsied sheep, a bluish-purple pigment was observed in the skin, subcutaneous tissue, fat, muscles, cartilage, bones, serous membranes of the forestomachs, kidneys, adrenal glands, and the mucosa of the uterus, urinary bladder, urethra, vagina, trachea, bronchi, and bronchioles. Some bone surfaces, the intima of large arteries, tendons, muscle insertions, and ligaments had a yellow-brown or light brown pigment. However, the pigment was not observed upon histologic examination of tissues, suggesting that the pigmentation is caused by a plant. Two plants, Rhamnidium molle and Pereskia bahiensis, were fed to experimental sheep and rabbits, but did not cause pigmentation.

Studies on the parasitic fauna of migratory sea birds of the Puffinus genus are scarce. Therefore, the aim of this study was to identify parasites of 16 specimens of Puffinus puffinus (Procellariiformes, Procellariidae) that died during the period of June 2011 to December 2011 at the Wildlife Screening Center (CETAS) of the Brazilian Institute of Environment and Natural Resources (IBAMA) in Cabedelo, Paraíba. During necropsy, biting lice and/or gastrointestinal helminths were collected in seven (43.7 %) birds. Lice were collected in five (31.2 %) birds, and the species identified were Halipeurus diversus, Trabeculus aviator, Austromenopon paululum), Saemundssonia sp. and Naubates sp. The prevalence of helminths was also 31.2 %. The nematodes species were Seuratia shipleyi and Contracaecum sp., and cestodes were Tetrabothrius sp. This is the first record in Brazil of Naubates sp., Seuratia shipleyi, Contracaecum sp., and Tetrabothrius sp. in Puffinus puffinus.

A survey on toxic plants for ruminants and equidae was performed on the municipalities of Juazeiro do Norte, Crato, Barbalha, and Missão Velha on the Ceará state, Northeastern Brazil. Twenty one interviews were realized with farmers, veterinary practitioners, agronomists and agriculture technicians. Poisonings by Ipomoea asarifolia mentioned in 38% and 19% interviews as toxic for bovines and sheep, respectively, and Enterolobium contotisiliquum, mentioned as toxic for cattle (47.6% of the interviews) and sheep (4.7%), were more frequent. Also occur in the region poisonings by Mascagnia rigida (38% of the interviews), Anadenanthera colubrina var. cebil (=A. macrocarpa) (14%), Ricinus communis (14%), Thiloa glaucocarpa (9%), and Sorghum halepense (4%) in cattle, Mimosa tenuiflora in cattle, sheep, and goats (38%), Brachiaria decumbens in sheep and cattle (38%), Manihot spp. in cattle and goats (28% ), and Leucaena leucocephala in sheep and horses (4%). Several plants previously unknown as toxic, but mentioned by the respondents as poisonous, were given to experimental animals at different doses. Only Casearia commersoniana was toxic to goats at the daily doses of 20g kg-1 body weight during 2-4 days. Clinical signs, similar to those reported by the farmers, were stiffness, mild bloat, polaquiuria, vocalization, jugular engorgement and pulsation, swaying gait, falling, spasticity, paddling movements, opisthotonos, and tachyicardia and dyspnea followed by bradycardia and bradypnea. Deaths occurred 6 and 19 hours after first clinical signs. No significant gross or histologic lesions were observed. It is concluded that poisonings by plants are important cause of losses in the region, which has a population of 53,473 bovines, 4,799 goats, 9,149 sheep, and 7,060 equidae.

The objective of this study was to describe clinical, laboratorial and histopathological findings of systemic phaeohyphomycosis in a dog. The animal was presented with a history of fatigue, ascites, lethargy, weight and appetite loss and polydipsia. The therapy for erliquiose and babesiosis was started. After 11 days, the dog returned with severe liver disease and died after forty-eight hours. At necropsy, there were black nodules in various abdominal organs. Histologically, necrotizing granulomatous reaction with hemorrhage associated with myriads of intralesional pigmented fungal hyphae was observed in multiple organs, characterizing systemic phaeohyphomycosis. Thus, it is important to include this illness in the differential diagnosis of severe liver diseases in dogs with ascites.

RESUMO

The aim of this research was to study the contribution of Toxoplasma gondii to reproductive failure using nested PCR and histopathological examination of fetuses, stillborns and placentas. We examined 245 organs of fetuses and 28 placentas from 35 abortions and stillborns from naturally occurring miscarriages in sheep in the State of Pernambuco, Brazil. At necropsy, fragments of brain, cerebellum, medulla, lung, heart, spleen, liver and placenta were taken for nested PCR and histopathological tests. Pathological examination revealed macroscopic lesions, suggesting T. gondii infection in 5/35 (14.3%) of the placentas. The histopathological examination revealed no lesions characteristic of toxoplasmosis in the organs investigated. In the five placentas, lesions consistent with toxoplasmosis were observed as an inflammatory non-suppurative infiltrate, along with multiple necrosis and mineralization. Nested PCR showed three aborted fetuses and two stillborns (14.3%) to test positive for T. gondii, with DNA amplification in all organs and the placenta, especially the heart and the placenta, which are the tissues of choice. This study substantiates the theory that T. gondii is involved in miscarriages and stillbirths and in the placentas of naturally infected sheep in Brazil. Such findings have not previously been described in the national literature.

A 12-year-old poodle dog was presented with a history of severe dyspnea. Radiology revealed diffuse pulmonary lesion which was diagnosed as interstitial pneumonia. At necropsy there were multiple small coalescent nodules distributed throughout the lung parenchyma. Histologically the nodules consisted of cubic to low columnar atypical epithelial cells which were PAS-negatives. In the immunohistochemistry reaction this cells were marked for cytokeratin, TTF (thyroid transcription factor)-1 and surfactant apoprotein A (SP-A). The neoplastic cells were negative for vimentin and cromagranine A. The necrotic areas were not marked for TTF-1 and SP-A. Based of the histopathological and immunohistochemistry findings a diagnosis of non-mucinous bronchioloalveolar carcinoma with diffuse involvement of the lung was made. This is a rare form of presentation for this tumor, and it can mimetize pneumonia on clinical and morphological examination. A definitive diagnosis for this condition should be based on histopathology and immunohistochemistry.

RESUMO

Pulmonary choristoma is a rare malformation reported in different animal species defined as a mass of normal histological pulmonary tissue in an abnormal location. A case of pulmonary choristoma and meningocele is reported in a calf that presented a fluctuating subcutaneous fluid containing mass, measuring 15 x 15 x 20cm in the skull frontal region. The skin covering the sac was surgically removed. Macroscopically, subcutaneous nodules up to 2cm in diameter with irregular whitish areas mixed with red areas were observed. In the histological examination, pulmonary lobules tissue composed by alveoli, bronchi, bronchioles and cartilage were observed. Dilated blood vessels and hemorrhages were present between the lobules. In this case the pulmonary choristoma was associated with meningocele, and probably was the mechanical cause for the failure of the skull closure.
